In order to clarify the effect of 2-furaldehyde on the cell membrane of Klebsiella pneumoniae, a membrane protein, 2-furaldehyde dehydrogenase was purified by QAE-Sephadex Q-50 anion exchange column chromatography and Sephadex G-100 gel filtration column chromatography. For the determination of the protein¢¥s molecular weight, nondenatured polyacrylamide gel electrophoresis was performed- the plotting method of [Log_(10)
KR(retardation coefficient) versus Log_(10) mol.wt.] showed a lowest experimental % Error deviation, and its molecular weight was estimated about 94,400 from the plot. And SDS-polyacrylamide gel electrophoresis revealed that 2-furaldehyde dehyrogenase had two nonidentical subunits, of which molecular `weight ratio for large subunit per small subunit was about 1.1.
