2-퓨르데알데하이드가 그램음성 세균인 Klebsiella pneumoniae 균주의 세포막에 미치는 영향을 규명하기 위하여 일차로 세포막에 존재하는 막단백질(膜蛋白質)의 특성을 살펴보고자 Klebsiella pneumoniae 균주의 세포막에 존재하는 막단백질인 2-퓨르데알데하이드 탈수소효소를 QAE-Se-phadex Q-50 음이온교환 원주크로마토그래피 및 Sephadex G-100 젤여과 원주크로마토그래피를 통하여 분리, 정제하였다. 다음 이 막단백질의 분자량을 측정하기 위하여 비변성 폴리아크릴 아마이드 젤 전기영동법이 사용되었고, 전기영동 결과 실험에 대한 % Error 편차가 가장 낮은 [Log_(10) K_®(지연 상수) 대 Log_(10) 분자량]함수를 통하여 분자량을 결정하였으며 이 때 산출된 2-퓨르데알데하이드 탈수소효소의 분자량은 약 94,400 정도 이었다. 한편 Sodium dodecyl sulfate 폴리아크릴아마이드 젤 전기영동결과 이 효소는 분자량이 서로 다른 두 개의 Subunit로 이루어졌음을 알 수 있었고, 이 두 Subunit의 분자량의 비, 즉 [Large subunit / Small subunit]의 값은 약 1.1이었다.
In order to clarify the effect of 2-furaldehyde on the cell membrane of Klebsiella pneumoniae, a membrane protein, 2-furaldehyde dehydrogenase was purified by QAE-Sephadex Q-50 anion exchange column chromatography and Sephadex G-100 gel filtration column chromatography. For the determination of the protein´s molecular weight, nondenatured polyacrylamide gel electrophoresis was performed- the plotting method of [Log_(10)
KR(retardation coefficient) versus Log_(10) mol.wt.] showed a lowest experimental % Error deviation, and its molecular weight was estimated about 94,400 from the plot. And SDS-polyacrylamide gel electrophoresis revealed that 2-furaldehyde dehyrogenase had two nonidentical subunits, of which molecular `weight ratio for large subunit per small subunit was about 1.1.
